Data & Biorepository

The focus of the Population & Data Sciences Core is to provide expertise for organizing cohorts of individuals with clinical and epidemiologic data as well as biospecimens; this will include support of studies of disease pathogenesis using advanced statistical and data sciences activities.

The Population & Data Sciences Core can provide access to cross-sectional and longitudinal clinical data and biospecimens from informative populations that include:

  • Individuals with clinical diagnoses of rheumatoid arthritis (RA). Subgroups include:

             - Seropositive RA, defined as anti-CCP and/or rheumatoid factor positive disease

             - Seronegative RA, defined as anti-CCP and rheumatoid factor negative

             - Early RA, defined as data and materials obtained from within 6 months of initial diagnosis of RA (some                          subjects are within days of diagnosis and have not used DMARDS).

             - Longstanding RA

  • Individuals who are at-risk for future RA. Subgroups include:

             - Anti-CCP+ individuals without clinical RA at baseline, some of whom later develop clinical RA

             - First-degree relatives of patients with RA (these FDRs may or may not have anti-CCP or rheumatoid                               factor positivity.

  • Autoantibody negative controls without RA
  • Inflammatory bowel disease (limited sample set)

 

Clinical, biomarker and biospecimens that can be available are as follows:

Clinical data:

Demographic features and environmental exposures (e.g. tobacco use, dietary intake), medical history and medication use, participant-reported joint symptoms, physician joint count (66/68), calculated disease activity measures, time-to-event (diagnosis of clinical RA or duration of follow-up).

 

Biomarker data:

  • Levels of anti-CCP, rheumatoid factor, C-reactive protein using clinical-grade assays (each time point)
  • HLA DRB1 shared epitope
  • Subset with fine specificities of anti-citrullinated protein antibodies 

Biospecimens:

Serum, plasma (EDTA and heparin), DNA (buffy coat), single PAXgene tube (whole blood RNA), PBMCs (in some cases), sputum, stool and urine.
